Reduced reserve is not yet an established ASH1L mechanism. It must be defined and measured as an altered perturbation threshold, response magnitude or coordination, response termination, recovery kinetics, or re-challenge behavior—not inferred from a difficult day or one treatment-linked change.

Who or what should be compared?

Patient-allele, corrected isogenic, dosage-series, and matched-reference cells in the same validated cell state, with independent clones, batches, challenge doses, and sham conditions.

STUDY DESIGN

How could the question be tested?

Use allele-aware, baseline-adjusted, time-resolved models across a safe, defined maturation step or tissue-appropriate challenge. Pre-specify dose, duration, sampling, viability, baseline state, peak, shutoff, recovery, re-challenge, correction, rescue, and negative controls.

What would support the proposed link?

a reproducible genotype × challenge × time effect after baseline adjustment is present, is not explained by toxicity or maturation mismatch, and moves toward control after correction or rescue.

What would weaken it?

the interaction disappears after baseline, viability, maturation, clone, or batch adjustment; the prespecified re-challenge pattern does not reproduce or cannot be separated from order or carryover effects; or the result does not show dose-appropriate attenuation under a validated correction or rescue condition.
