10Which biomarkers and outcomes measure meaningful change?

No biomarker is qualified to diagnose ASH1L-related disorder, track disease state, predict trajectory, or measure treatment response. A statistically responsive measure may still be unreliable, developmentally inappropriate, or unrelated to what matters to the person.

Who or what should be compared?

Predefined age and ability bands with sufficient repeated measurements to estimate reliability and change; analytic validation samples, natural-history samples, and external confirmation samples remain distinct.

STUDY DESIGN

How could the question be tested?

Embed analytic validity, test–retest reliability, age and ability feasibility, floor and ceiling effects, minimal detectable change, practice effects, rater effects, longitudinal sensitivity, and patient- or caregiver-defined meaningfulness within prospective natural history.

EVIDENCE GATE

What would support the proposed link?

the measure is reliable within its intended age and ability range, changes beyond measurement error, relates to a meaningful functional domain, performs in an independent sample, and has a prospectively defined interpretation.

STOP RULE

What would weaken it?

the measure is technically unstable, dominated by age or ability floors and ceilings, sensitive only to practice or rater, unrelated to meaningful function, or lacks an interpretable change threshold.
